چکیده

A ctin depolymerization is a death trigger for poppy pollen, according to Thomas et al. (page 221), that prevents self-fertilization. Some plants prefer not to be fertilized by pollen grains that are genetically similar to themselves. These plants express self-incompatibility (S) proteins. A match between the pistil’s S-proteins and the pollen’s S-allele triggers a rapid calcium infl ux, actin fi lament destruction, and pollen cell death. Several years ago, the researchers noticed that the amount of actin depolymerization far exceeded that required to inhibit pollen tube growth. The persistent actin destabilization also resembled that associated with animal cells about to undergo programmed cell death (PCD). The authors now fi nd that blocking actin depolymerization prevents pollen cell death in selfincompatible plants. Inducing actin depolymerization, on the other hand, activated pollen cell death. The dying cells showed the same pattern of DNA fragmentation and caspase-like activity as did pollen killed by self-incompatibility. Changes in actin dynamics have been linked to PCD in animal and yeast cells, and a variety of noxious external stimuli triggers actin rearrangement in plants. Actin may thus serve as a sensor for extreme environmental stress to initiate PCD or other changes in cell behavior. As even a short exposure to depolymerizing drugs killed pollen cells, a brief change in actin dynamics seems to be all that is needed to kick off signaling cascades.

Alterations in the actin cytoskeleton of pollen tubes are induced by the self-incompatibility reaction in Papaver rhoeas.

Self-incompatibility (SI) is a genetically controlled process used to prevent self-pollination. In Papaver rhoeas, the induction of SI is triggered by a Ca(2)+-dependent signaling pathway that results in the rapid and S allele-specific inhibition of pollen tube tip growth. Tip growth of cells is dependent on a functioning actin cytoskeleton. We have investigated the effect of self-incompatibili...

Actin-binding proteins implicated in the formation of the punctate actin foci stimulated by the self-incompatibility response in Papaver.

The actin cytoskeleton is a key target for signaling networks and plays a central role in translating signals into cellular responses in eukaryotic cells. Self-incompatibility (SI) is an important mechanism responsible for preventing self-fertilization. The SI system of Papaver rhoeas pollen involves a Ca(2+)-dependent signaling network, including massive actin depolymerization as one of the ea...

Microtubules are a target for self-incompatibility signaling in Papaver pollen.

Perception and integration of signals into responses is of crucial importance to cells. Both the actin and microtubule cytoskeleton are known to play a role in mediating diverse stimulus responses. Self-incompatibility (SI) is an important mechanism to prevent self-fertilization. SI in Papaver rhoeas triggers a Ca(2+)-dependent signaling network to trigger programmed cell death (PCD), providing...

Signal-mediated depolymerization of actin in pollen during the self-incompatibility response.

Signal perception and the integration of signals into networks that effect cellular changes is essential for all cells. The self-incompatibility (SI) response in field poppy pollen triggers a Ca(2+)-dependent signaling cascade that results in the inhibition of incompatible pollen. SI also stimulates dramatic alterations in the actin cytoskeleton. By measuring the amount of filamentous (F-) acti...

Proteins implicated in mediating self-incompatibility-induced alterations to the actin cytoskeleton of Papaver pollen.

BACKGROUND AND AIMS Sexual reproduction in angiosperms involves a network of signalling and interactions between pollen and pistil. To promote out-breeding, an additional layer of interactions, involving self-incompatibility (SI), is used to prevent self-fertilization. SI is generally controlled by the S-locus, and comprises allelic pollen and pistil S-determinants. This provides the basis of r...
